Comparison of different phosphorus-containing ligands complexing 68Ga for PET-imaging of bone metabolism

2010

Abstract 99mTc-phosphonate structures are well established tracers for bone tumour imaging. Our objective was to investigate different 68Ga-labelled phosphonate ligands concerning labelling kinetics, binding to hydroxyapatite and bone imaging using μ-PET. Seven macrocyclic phosphorus-containing ligands and EDTMP were labelled in nanomolar scale with n.c.a. 68Ga in Na-HEPES buffer at pH∼4. Except for DOTP, all ligands were labelled with >92% yield. Binding of the 68Ga-ligand complexes on hydroxyapatite was analysed to evaluate the effect of the number of the phosphorus acid groups on adsorption parameters. Adsorption of 68Ga-EDTMP and 68Ga-DOTP was >83%. For the 68Ga-NOTA-phosphonates …

Synthesis, Characterisation and Reactivity towards PdII and PtII of ortho-, meta- and para-Xylyl-Based Phosphorus-Containing Macrocycles

2002

The new meta- and para-xylyl-based phosphadithiamacrocycles 6-phenyl-6-phospha-3,9-dithiabicyclo[9,3,1]pentadeca-1(15),12,14-triene (L2) and 6-phenyl-6-phospha-3,9-dithiabicyclo[9,2,2]pentadeca-1(13),12,14-triene (L3) have been synthesised and characterised. The reactivity of these ligands and the previously described ortho-xylyl macrocycle 6-phenyl-6-phospha-3,9-dithiabicyclo[9,4,0]pentadeca-1(11), 12,14-triene (L1) with [PdCl2(cod)] and K2[PtCl4] has been studied. [MCl2(L)] complexes have been obtained and characterised by NMR spectroscopy. X-ray structural analyses have been carried out in the case of [PdCl2L2]·CH3CN (2·CH3CN) and [PdCl2L3] (3). In both compounds the PdII ion adopts a sl…

Unusual activity pattern of leucine aminopeptidase inhibitors based on phosphorus containing derivatives of methionine and norleucine

2010

Ligands containing bulky aliphatic P1 residues exhibit a high affinity towards cytosolic leucine aminopeptidase, a bizinc protease of biomedical significance. According to this specificity, a series of phosphonic and phosphinic compounds have been put forward as novel putative inhibitors of the enzyme. These phosphonic and phosphinic compounds were derivatives of methionine and norleucine as both single amino acids and dipeptides. The designed inhibitors were synthesised and tested towards the peptidase isolated from porcine kidneys using an improved separation procedure affording superior homogeneity. Unexpectedly, organophosphorus derivatives of methionine and norleucine exhibited moderat…

Speciation of chitosan-phosphate and chitosan-nucleotide systems in an NaCl aqueous solution

2010

AbstractThe speciation of chitosan (310 kDa) with organic (adenosine 5’-monophosphate, AMP, and adenosine 5’-triphosphate, ATP), and inorganic phosphorus containing ligands (phosphate and pyrophosphate) was investigated in NaCl aqueous solutions at I = 0.1mol L−1 and T = 25°C. For all the systems, the investigated results obtained gave evidence for the formation of (chitosan)LHi complex species (L = nucleotides, phosphate and pyrophosphate; i = 1 to 4, but for AMP, i = 1 to 3). The stability data of complex species were used to calculate the sequestering ability of chitosan towards phosphorus compounds considered here, expressed as pL50 i.e., – log(total chitosan concentration) necessary to…
